There are actually two no-fly lists. The one maintained by the DHS which takes a Federal Judge to get off of, and one maintained by each Airlines (referred to as the no boarding list) which is actually damn near impossible to get off of.

I've always wanted to to do this on a plane. Not while in flight, but definitely while on the ground. Thing is though, I know better than to fuck with airplane safety equipment outside of a legitimate emergency.
