Mean lethal dose for mice is about 25mg./kg, rats 50mg/kg, dogs 80 mg./kg.

In rhesus monkeys daily for 16 days at 1.2 mg/kg, was tolerated well.

In humans doses up to 2 mg/kg single dose were tolerated with mild to moderate side effect in studies. Also at 1mg./kg for 3 times in a week and at 1.6mg/kg 2x a week for 12 weeks. These are pretty high doses.

Some overdoses in humans in the range of 15 mg./kg have recovered. People who get that much start drooling and twitching and are heavily sedated, because it can fuck with your CNS. The drug has a hard time crossing the blood brain barrier, which is largely made up of p-glycoprotein. Invertebrates like parasites dont have a BBB and are susceptible in very low concentrations, but not mammals. Theoretically 1 in a million people may have a sensitivity due to mutation of the MDR gene, like some herding dogs do who cant take this drug. Some drugs which affect p-glycoproteins could lower your tolerance but at prescribed doses this is rarely observed. See the FLCCC site for recommendations. They are using drugs with some mild interactions with IVM together with it , such as glucocorticoids, lipitor, aspirin. Observed interactions are rare.

To put these doses in to perspective, the FLCCC is recommending 0.4 to 0.6 mg./kg for 5 days. For a 150 lbs person that is 27 to 40 mg a day x 5, cumulatively 2 to 3 mg /kg , or about 2 tubes of ivermectin horse paste for 1250lbs horse maximum cum.
